Métodos de muestreo
Each community was surveyed with a particular sampling method. The fitoplancton was collected for both qualitative and quantitative analysis. The sampling for the qualitative analysis was the following: Samples taken with a plancton net of 18 um pore which were kept on two 60 ml plastic containers. The first container (live sample) was preserved cold till its analysis on the lab. The content of the second container was fixed with 1% formaldehyde The sampling for the quantitative analysis was the following: One single 100ml water sample kept in a container with a 2% lugol solution. The zooplancton was collected through water filtering with a 55 um mesh making 10 drags per water body, with samples being kept on a solution of 4% formaldehyde. The fitobentos and perifiton were sampled by taking 3 substrate samples from each sampling station. For some macrophytes, the sampling was made by taking a significant portion of them. The samples were integrated in a 100-150 ml container and were kept in a dark and cold place. The macroinvertebrates were sampled by using a Surber, with a polystyrene 500 um mesh width and a sampling surface of 0,1 m2. In lotic environments the mesh was placed against the current and the area taken by the mesh was removed (0,1 m2) during 1 minute. In lenthic environments, such as the lagoons, a 290 um hand net to do 10 drags of 1 m length in total. The samples were kept in air tight containers fixed on 80% alcohol.
| Área de Estudio | The study was developed as part of the second phase of Total Austral´s Biodiversity Management Plan for their operational sites in the Cuenca Marina Austral- 1 on-shore operational area. The phase is called Biodiversity Action Plan Design. In this phase, the consultant does and environmental baseline in order to collect reliable scientific data on the Biodiversity status of the site. This is made with the objective of later on design appropriate biodiversity conservation actions to be implemented in the site. This study is a Limnology survey campaign done during the winter season in several freshwater and saltwater environments present in CMA-1. These environments were detected previously in the Critical Habitats and Key Biodiversity Features campaign. There were 12 sampling stations: Hito 1 Lagoon, Alfa River, Beta river, Cullen Ríver (two sampling points), Cullen River estuary (2 sampling points) and interior lagoons (3 sampling points) However, no samples could be collected from Alfa estuary and Beta estuary, because there was no water on them during the survey. The water body from which each observation comes is specified in the "parentEventId" column, following this abbreviation criteria: LH1#1 = Hito 1 Lagoon, Sample 1 RA#1 = Alfa River, Sample 1 RB#1 = Beta River, Sample 1 CD#1 = Distal Cullen River, Sample 1 CP#1 = Proximal Río Cullen, sample 1 EC#1 = Cullen estuary, sample 1 EC#2 = Cullen estuary, sample 2 LI#1 = Interior Lagoon 1 LI#2 = Interior Lagoon 2 LI#3 = Interior Lagoon 3 This campaign included surveying the fitoplancton, zooplancton, fitobentos, perifiton and macroinvertebrates communities, as well as measuring physiochemical parameters from each water body surveyed. The campaign was developed from the 14th to the 21th November 2024. |

| Control de Calidad | The identification was made following this criteria: The fitoplancton and zooplancton identification was done at specific level, by the Limnology Laboratory of the Cento Austral de Investigaciones Científicas (CADIC-CONICET). Fitobentos y perifiton: The separation and identification of organisms was made by using a stereoscopic magnifying glass. Macroinvertebrates: In each of the sites, at least 3 replicas were made- The taxonomic determinations were made to the lowest taxonomic level possible, by consulting dichotomic keys. Some of the ones used were Miserendino & Pizzolón (1999), Miserendino (2009), Fernández & Domínguez (2009), Palma, (2013) or another more updated bibliography. |
Descripción de la metodología paso a paso:
- Once the sampling was made in each sampling station, some quantitative analysis were made. The zooplancton countings were made with a 1 ml capacity counting camera, using a binocular microscope to calculate the number of individuals per litre. All algae present were counted using an inverted microscope. The macroinvertebrates were easliy counted by hand.
